چکیده

Skeletal regeneration is accomplished by a cascade of biologic processes that may include differentiation of pluripotential tissue, endochondral ossification, and bone remodeling. It has been shown that all these processes are influenced strongly by the local tissue mechanical loading history. This article reviews some of the mechanobiologic principles that are thought to guide the differentiation of mesenchymal tissue into bone, cartilage, or fibrous tissue during the initial phase of regeneration. Cyclic motion and the associated shear stresses cause cell proliferation and the production of a large callus in the early phases of fracture healing. For intermittently imposed loading in the regenerating tissue: (1) direct intramembranous bone formation is permitted in areas of low stress and strain; (2) low to moderate magnitudes of tensile strain and hydrostatic tensile stress may stimulate intramembranous ossification; (3) poor vascularity can promote chondrogenesis in an otherwise osteogenic environment; (4) hydrostatic compressive stress is a stimulus for

Skeletal regeneration in amphibians.

T H E failure of de novo regeneration of extirpated bones in adult urodeles has been described repeatedly. Wendelstadt (1901) reported the absence of skeletal regeneration following the removal of either the ulna or radius from the otherwise intact limbs of salamanders. Reed (1903) and Morrill (1918) likewise failed to obtain regeneration of previously excised fibulae in salamander hind limbs. ...

Plasticity and recovery of skeletal muscle satellite cells during limb regeneration.

Salamander limb regeneration depends on local progenitors whose progeny are recruited to the new limb. We previously identified a Pax7(+) cell population in skeletal muscle whose progeny have the potential to contribute to the regenerating limb. However, the plasticity of individual Pax7(+) cells, as well as their recovery within the new limb, was unclear. Here, we show that Pax7(+) cells remai...

G-CSF influences mouse skeletal muscle development and regeneration by stimulating myoblast proliferation

After skeletal muscle injury, neutrophils, monocytes, and macrophages infiltrate the damaged area; this is followed by rapid proliferation of myoblasts derived from muscle stem cells (also called satellite cells). Although it is known that inflammation triggers skeletal muscle regeneration, the underlying molecular mechanisms remain incompletely understood. In this study, we show that granulocy...

The effects of obesity on skeletal muscle regeneration

Obesity and metabolic disorders such as type 2 diabetes mellitus are accompanied by increased lipid deposition in adipose and non-adipose tissues including liver, pancreas, heart and skeletal muscle. Recent publications report impaired regenerative capacity of skeletal muscle following injury in obese mice. Although muscle regeneration has not been thoroughly studied in obese and type 2 diabeti...

Muscle Regeneration and Compensatory Growth

Regeneration of skeletal muscle has been documented from an anatomical perspective for a number of years. In recent times the mechanisms that underline skeletal muscle regeneration have been extensively investigated and the topic of a comprehensive edited review by Mauro ( I 979). This brief review wil l focus on cellular proliferation aspects of skeletal muscle regeneration and wil l not attem...

Smad4 restricts differentiation to promote expansion of satellite cell derived progenitors during skeletal muscle regeneration

Skeletal muscle regenerative potential declines with age, in part due to deficiencies in resident stem cells (satellite cells, SCs) and derived myogenic progenitors (MPs); however, the factors responsible for this decline remain obscure. TGFβ superfamily signaling is an inhibitor of myogenic differentiation, with elevated activity in aged skeletal muscle. Surprisingly, we find reduced expressio...
